DUALITYBIO-B Inks Landmark Global ADC Collaboration with Roche's Genentech Using Proprietary DUPAC Platform

DUALITYBIO-B (09606) has announced the signing of a collaboration and license agreement with Genentech, Inc., a member of the Roche Group, dated August 27, 2026. The partnership leverages DUALITYBIO-B's proprietary DUPAC platform to develop next-generation antibody-drug conjugates (ADCs) targeting multiple oncology indications.

Under the terms of the agreement, DUALITYBIO-B will spearhead the discovery phase and early-stage global clinical development of the collaborative projects. Genentech will hold an exclusive worldwide license to any ADCs resulting from the collaboration, and upon the completion of Phase 1a clinical trials, Genentech will assume full responsibility for subsequent clinical development and commercialization efforts.

The financial structure of the deal provides DUALITYBIO-B with an upfront payment of $45 million, alongside the potential to receive aggregate milestone payments exceeding $1 billion across all collaboration programs, contingent upon achieving specified development, regulatory, and commercial milestones. Additionally, DUALITYBIO-B is entitled to tiered royalties based on annual net sales of any approved products derived from the collaboration.

The board of directors believes this collaboration enables DUALITYBIO-B to address the growing unmet medical needs of oncology patients whose tumors exhibit resistance or refractoriness to the payload classes utilized by currently approved ADCs. The company has developed DUPAC, a novel family of mechanistic payloads engineered to overcome payload resistance, thereby addressing these challenges and allowing each ADC program to explore a broader spectrum of relevant tumor biology.

By integrating DUALITYBIO-B's payload innovation and early global clinical development capabilities with Genentech's profound oncology expertise and worldwide development reach, this collaboration aims to advance programs with greater speed and scale, ultimately delivering differentiated medicines to more patients globally.

DUPAC represents DUALITYBIO-B's proprietary ADC payload platform featuring a novel mechanism of action, and it stands as one of four ADC technology platforms developed by the company, alongside DITAC, DIMAC, and DIBAC. The DUPAC platform encompasses multiple payloads, including DUP5, DUP9, and DUP10, each operating through distinct anti-tumor mechanisms.

Preclinical data for DUP5-based ADCs and DUP9-based ADCs, including activity in tumor models relatively insensitive to topoisomerase inhibitor payloads and tolerability in non-human primates, have been presented at several medical conferences, including AACR 2025 (Abstract 5454), the AACR-NCI-EORTC International Conference 2025 (Abstracts B129 and B130), and AACR 2026 (Abstract 2657).
